Hij debuteerde in de Formule 1 bij Williams tijdens het seizoen 2017 en reed daar tot 2018. Vervolgens stapte hij in 2019 over naar Racing Point en bleef bij het team toen het werd omgedoopt naar Aston Martin. Het team wordt mede-eigendom van zijn vader, Lawrence Stroll. Gedurende zijn carrière behaalde Lance drie podiumfinishes: de derde plaats in de Grote Prijs van Azerbeidzjan 2017, de Grote Prijs van Italië 2020 en de Grote Prijs van Sakhir 2020.
In 2023 behaalde Stroll zijn beste resultaat ooit in de coureursstand, met een 10e plaats. Toch was hij nog steeds geen partij voor zijn teamgenoot Fernando Alonso, die hem consequent overtrof.
De prestaties van Aston Martin namen af in 2024, en de situatie verslechterde ook voor Stroll. Weer slaagde hij er niet in om Alonso’s snelheid en resultaten bij te benen.
